GENETIC DIAGNOSIS OF THE BACTERIAL ISOLATES FOR PESTICIDE ANALYSIS OF ACETAMIPRID AND STUDY OPTIMAL CONDITION FOR THEIR GROWTH

Abstract

This study aimed to determine the most effective bacterial species for breaking down acetamiprid in soil samples obtained from the greenhouses, 10 bacterial isolates were collected from soil contaminated with pesticides taken from greenhouses of the Horticulture Department/Ministry of Agriculture (Abu Ghraib,Iraq) which has been using these pesticides for a long time on a wide range of agricultural crops such as cucumbers, tomatoes, eggplants and squash. The best bacterial isolates that showed the ability to decompose the pesticide with high efficiency for 21 days are isolates 7, 8, 9, and 10. Isolate No. 9 which had the best biodegradation of the pesticide, was identified and thus genetically diagnosed. The results of the genetic diagnosis of bacteria No. 9 showed that the local bacterial isolate belongs to the genus Lysinibacillus of the bacterial strain Lysinibacillus fusiformis, which was registered as the first local isolate in the gene bank that worked to degrade or break down the pesticide.The optimum conditions for the growth of the isolated and most efficient degrading bacteria in the decomposition of the pesticide were determined. The optimum temperature for the growth of the strain Lysinibacillus fusiformis (PQ109093) was 28 C˚,and the optimum pH for growth was 7.0,and the optimum concentration for the decomposition of the pesticide by bacteria was 25,50 ppm at the concentrations used for the pesticide.
